Jul 04, 2019 · Pressure cook the liquid culture. Add the jars to your pressure cooker and bring them to 15 PSI for approximately 15-20 minutes. Read the instructions on your pressure cooker and always remember to fill it above the base plate with water. Leave the culture to cool to room temperature.
Millex-GP filters are recommended for sterile filtering protein solutions, tissue culture media, buffers and additives. Millex filters with Durapore® (PVDF) membrane are the lowest protein-binding syringe filters available. Use them to filter protein solutions. The 0.2 µm filters are sterilizing grade.
It is important that the liquid is below 106 degrees before inoculating with your liquid cultures. Anything at or above that temperature will kill your mushrooms. Shake the syringe you bought with the culture in it till the strands are broken up and loose. Then inject about 3cc into the jar.
For samples composed of acids and bases, hydrophilic PTFE membrane syringe filters and PES membrane syringe filters may be considered. For samples composed of nonaqueous solvents that are not compatible with nylon, PTFE membrane syringe filters and polypropylene membrane syringe filters may be considered.
